The Potential of Treated Palm Oil Mill Effluent (Pome) Sludge as an Organic Fertilizer

Palm oil mill contributed a significant benefit to agro-based industry and social-economic for Malaysia. Palm oil mill effluent (POME) is considered as a polluted wastewater and the treated POME sludge was produced from the open treatment ponds. The objective of this study was to determine the physicochemical characteristics of treated POME sludge and its potential as an organic fertilizer. It was collected from the dumping ponds in Felda Jengka 8, palm oil mill. Physicochemical characteristics, sampling and preparation of samples were analyzed according to the standard method of soil and the wastewater. The samples were collected after one and six month of age with different depths (one, two and three meters). The statistical analysis revealed that the depth was not significant on the physicochemical characteristics. The characteristics of the treated POME sludge was measures using CHNS-O, C/N ratio, solid analysis, heavy metal, macro and micronutrient, moisture content, and pH. However, the elements of oxygen, iron and pH were shown an interaction effects with time. In conclusion, the treated POME sludge has shown significant effect and the potential used as an organic fertilizer. Indeed, further studies on crops response are being conducted to prove the findings.

Growth and Yield of Eggplant (Solanum Melongena L.) on Various Combinations of N-source and Number of Main Branch

This research was conducted to increase production of eggplants through combination of N-source and number of main branch has done onthe field of andosol in Poncokusumo - Malang, 600 m asl, pH 5.4, from August to December 2013. The experiment used a Randomized Complete Block Design with 2 factors and 3 replications. Factor 1 was proportion of inorganic - organic N fertilizer (138 kg N ha-1): 100% Urea, 75% Urea + 25% goat manure, 50% Urea + 50% goat manure, and 25% Urea + 75% goat manure. Factor 2 was number of main branches: 1, 2 and 3 main branches. Results showed that there was no interaction effect between treatment combinations of organic-inorganic sources of N and the number of main branches to all observed variables. Treatment using the combination of 75% Urea + 25% goat manure increased the plant growth and gave the highest fruit yield (49.20 t ha-1) in comparison with combination using other fertilizers and 100% Urea. The lowest was derived from the application of 100% Urea, 35.61 t ha-1. Cultivation of eggplant with 3 main branches has resulted better growth and fruit yield than 1 and 2 main branches, 50.85, 47.91 and 30.79 t ha-1, respectively.

Efficient Strategies for Elimination of Phenolic Compounds During DNA Extraction From Roots of Pistacia Vera L.

Optimization of DNA extraction protocols for plant tissues and including endophytic microorganisms is a critical step of advanced plant-microbe interaction in agricultural studies. Pistachio (Pistacia vera L.) root tissue contains high levels of polyphenols have been known as major extract contaminants and inhibitors of enzymatic activities during amplification. The present study aimed to develop reliable strategies to purify DNA from Pistachio root samples. Inhibiting substances were removed from DNA through a process including extraction with hot detergent contains SDS-Tris- EDTA, AlNH4(SO4)2.12H2O as chemical coagulating factor and CTAB-NaCl. Following typically organic extraction/alcohol precipitation, denaturing agarose electrophoresis performed to purify probable remain contaminants. The purified DNA was enough free of polyphenols based upon loss of color and spectral quality (260/230>1.6) and efficiently amplified during polymerase chain reaction particularly in the present of GC-clamp primers. This method proved well with detection of Glomus sp. (arbuscular mycorrhiza fungi) associated with Pistacia vera L. using denaturing gradient gel electrophoresis (DGGE).
