Investigating Effects of Grape Seed Extract on Neuropathic Pain in the Streptozotocin-induced Diabetic Mice.

Diabetes mellitus is a complicated and serious health problem involving peripheral neuropathy. This situation causes to loss of senses, tingle and pain. Diabetic peripheral neuropathy (DPN) affects 236 million people around the World. Hence there is a need to investigate alternative ways of cure focusing on DPN. Flavonoids have potential on pain due to their permeability characteristic in the capillary microcirculation system and in lowering blood pressure. Flavonoids are common in grape seed. The main flavonoids of grape seed involve proanthocyanidins which might be an effective agent in cure of pain in DPN. The purpose of this study is to investigate effects of grape seed extract on neuropathic pain in the Streptozotocin-induced diabetic mice. In the study, 50 eight-week old BALB-C strain mice in five different groups (Control, Diabetic Control, Control+25 mg/kg, Diabetes+25 mg/kg and Diabetes+50 mg/kg) were used. To induce diabetes in thirty of these animals, single dose Streptozotocin (180 mg/kg) was administered intraperitoneally. After diabetes was observed, grape seed extract (25 mg/kg and 50 mg/kg body weight) was administered by oral gavage in three groups (Cont­rol+25 mg/kg, Diabetes+25 mg/kg and Diabetes+50 mg/kg) during six weeks. At the end of the second and sixth weeks, pain threshold measurements in hot plate test were performed in line with a predetermined thermal pain model. Also the tissues of the sciatic nerve and abdominal aorta from the animals were histologically investigated. As a result hot plate measurements, pain threshold values of the animals in Diabetes+50mg/kg group significantly differed from the measurements of the animals in control group in the first measurements and from the animals in Diabetes+25 mg/kg group in the second measurements (p<0.05). However pain threshold values of the animals in Diabetes+25 mg/kg group differed significantly from the values of the animals in control+25 mg/kg group and control group. It means pain threshold values of the animals in Diabetes+50 mg/kg and Diabetes+25 mg/kg groups were significantly lower than the values of the animals in the other groups. The results of histological investigations showed that degenerations of myelin sheet and axons in diabetic control group were decreased significantly in Diabetes+50 mg/kg and Diabetes+25 mg/kg groups. Moreover degenerations of aorta tissues of animals in diabetic group were not seen in the animals of Dia­betes+50 mg/kg except for tunica adventitia inflammation. It can be said that grape seed extract decreased threshold of neuro­pathic pain in the Streptozotocin-induced diabetic mice and prevented degenerations of myelin sheet and axons, and aorta tissues.

Synergy of Piracetam and Aminalon in the Cerebroprotective Effect of Olatropyl.

Optimization of pharmacotherapy of neurological and psychosomatic diseases led to the creation of com­bined medicines containing two or more components with different mechanisms of action, differently affecting the pathogenesis and clinical manifestations of specific forms of pathology. Combined drugs that help normalize the central regulatory mechanisms of the development of somatic pathology, improve brain metabolism and hemodynamics, stabilize the cortical-subcortical relationships, the disorders of which lie at the base of many forms of neurological and psychosomatic pathology have been particularly popular in recent years. In this article we propose options for opti­mizing nootropic therapy with olatropyl, which is a combination of two "true" nootropics - gamma-aminobutyric acid (aminalone) and piracetam in one drug form. Such a combination allows one-stage implementation of a complex of multifaceted and diverse effects on the central nervous system characteristic of both piracetam and aminalone, but achieved through fundamentally different mechanisms of action of these agents. The mechanism of action of olatropil demonstrates the pronounced synergism of the monocomponents of the preparation. This combination allows to reduce the therapeutic dosages of each of the active substances by 2 times, which, in turn, leads to a decrease in the incidence and severity of possible side effects; leveling the exciting effect of piracetam and providing higher safety and efficacy. The drug is an effective tool for the therapy of various forms of cerebrovascular pathology, improving cognitive functions, mental activity, psychoemotional status, autonomic functions, favorably affecting the quality of life of patients. The uniqueness of the mechanism of action of olatropil, consisting in the occurrence of the fact of pharmacodynamic synergy in the combined use of piracetam and aminalone in a single dosage form, contributes to the enhancement of positive pharmacological effects and the leveling of a number of negative elements of pharmacodynamics of preparation constituents.

L-lysine Escinat, Thiotriazolin, Gordox and Mydocalm Influence on Oxygen Tension in the Intestinal Wall and Acid-base Balance and Limited Proteolysis in Intestinal Venous Blood in Terms of Intraabdominal Hypertension Modeling.

In acute experiments on rabbits there were studied changes in oxygen tension in the intestinal wall tissues, acid-base balance and limited proteolysis and its inhibitors in intestinal venous blood, protective action of L-lysine escinat (0,15 mg/kg / single dose), thiotriazolin (25 mg/kg / single dose), aprotinin (gordox) (10,000 units/kg / single dose) in sequential modeling of standard levels increasing of intra-abdominal hypertension (IAH) — 50, 100, 150, 200, 250, 300, 350 m H2O, and also of tolperison (mydocalm) (5 mg/kg / single dose) on modeling of stable 3-hour IAH 200 m H2O. The IAH modeling was performed by means of stand of our construction. Under the influence of IAH the compensated metabolic acidosis in intestinal venous blood with a compensative hyperpnoe develops, decline of oxygen tension in tissues and activating of a limited proteolysis as well as decline of its inhibitors activity in intestinal venous blood occur. By the degree of metabolic acidosis prevention investigational preparations were distributed as follows gordox > thiotriazolin = L-lysine escinat = mydocalm, and by prevention of decline of oxygen tension in tissues — thiotriazolin > L-lysine escinat > mydocalm > gordox, it is is connected with different rate of methabolic products excretion into the blood, due to the influence on blood circulation and transcapilary exchange. By the degree of prevention of proteolytic activity and inhibitory potential changes, investigational preparations were distributed as follows: gordox > mydocalm > thiotriazolin > L-lysine escinat, this is connected with inhibition of proteolysis in gordox, and in other ones – with reduction of ischemic damage of tissues. Owing to different mechanism of action thiotriazolin, L-lysine escinat and mydocalm may be simultaneously recommended for a conservative treatment of patients with intraabdominal hypertension syndrome.
